While the analytical public agreed on the chromatographic method for determination of iso-alpha acids in beer (the EBC 7.8. method is used in international trials) there is no obligatory method for extraction these compounds and every laboratory uses its own procedure. Beside the classical liquid-liquid extraction many different ways are used to isolate target compounds, especially the SPE. Unlike the reverse phases used based on modified silicagel, a new polymeric sorbent STRATA X exceeds by higher capacity and versatility has been tried. In this study was monitored impact of collected yeast washing intenzity and storage condition on its quality.We worked with three lager brewing yeast strains No. 2, 7, 95 (RIBM 655 - Culture Collection of Brewing Yeasts) with different genetic properties. Yeast viability was determined partly by methylene blue staining, partly by propidium iodide staining with flow cytometric analysis. Both staning methods were compared.
