Extraction colorimetric method based the biuret reaction can be used to advantage for rough determination of protein content in barley, since its gives reasonably reliable results. Its accuracy depends in the standardization of samples and fineness of grinding. The original method developed by Johnson and Craney has been adapted to meet concrete, specific conditions. Isopropyl alcohol is now used as an extracting agent, basic salt is used instead of cupric carbonate, and vacuum filtration has been replaced with forced pressure membrane filtration. The method is suitable for routine analyses carried out to determine protein in barley.
