A procedure and conditions for the determination of the volume of conical bottom fermenting vessels for the single-tank beer production are described in the article. Examples of the necessary capacity are given with respect to a building of new factories and an enlargement of contemporary capacities. The introductory part of this work presents information on the utilization of the Yeast Collection of the Research Institute for viticulture and Enology in Bratislava and on the production of liquid yeast starters. Selected wine yeasts strains used in Czechoslovak wine industry are briefly characterized. Pectinase complexes of commercial preparations Pectinex Ultra (Schweizerische Ferment AG, Basel) and Leozym (Slovlik, n. p., Leopoldov, Czechoslovakia) containing endopolygalacturonase, exopolygalacturonase, pectin, lyase, and pectinesterase were irreversibly immobilized by adsorption on polyethyleneterephthalate - orsilen )Silona, n. p. The immobilization resulted in a lowering of pectinolytic activity; the measure of the lowering depended on the amount of the enzyme fixed on the carrier. A method for determination of the volumetric oxygen transfer coefficient KLa from dynamic measurement data under conditions of both excess oxygen and oxygen limited growth of microorganisms is described. The KLa determination follows from the integrated form of the oxygen balance equation and involves a correction for the actual cultivation conditions during measurement.
