Ergosterol in brewing yeasts.Peer-reviewed article

An information study of ergosterol content in brewing and baker's yeasts was made. Six methods for the ergosterol determination were tested. As a result the Longley method was used in further work. When a cell disintegration was made prior to the cell hydrolysis a higher ergosterol yield was found. Steroids were isolated from the cell mass by the alkaline hydrolysis with a following extraction by cyclohexane. The ergosterol content in bakers yeast was about 2 to 5 times higher than that in brewing yeasts. An application of modern computer techniques in the sale department of each brewery increases the quality of commercial transactions of the whole trust. For our production conditions the microcomputer Robotron 1715 from GDR was chosen. This computer will treat all the orders including their changes and on a base of the production possibilities also will perform the bills of delivery. The polarographic method for the determination of the free sulphur dioxide content in wines was developed. The method is quick, correct, enables to make a record and is suitable for the inspection of the free sulphur dioxide level both in white and red wines.(In Slovak, English summary only)

Productions of 10 g.l-1 citruline and 30 g.l-1 ornithine were achieved during 96 hours submerged cultivation of auxotrophic mutants of Corynebacterim sp. 9366 which were dependent to arginine. The ornithine preparation process by a fermentation has been developed. The production of 30 g.l-1 ornithine was achieved after 60 hours of the cultivation. The production medium contained molasses and hydrolyzate of peanut meal.(In Czech, English summary only)

The following types of bioreactors are described: reactor with a turbine impeller, loop reactors with a marine propeller or with combined impellers, various types of systems without gas spargers and reactors with multiple impellers. The principle, the maximum vessel volume and possible applications are discussed by each type of the reactor. With respect to the keeping of the process asepticity possible ways of the scale-up with respect to the reactor shape are mentioned.(In Czech, English summary only)

The cooling circuit of the 200 m3fermenter consists of centrifugal pumps (Sigma, tape NFT) and plate exchangers (Maschine Works Chotěboř). The best function of this circuit was achieved when the medium was taken of from the place with the lowest gas hold-up in the fermenter. The pump function and the broth discharge rate is constant with the hold-up of air up to 35 %. Under these conditions the pressure loss of the plate exchanger remains up to 0,15 MPa at he flow rate of 300 m3.h-1.
