The article deals with the role of proline in malting and brewing technology and with the specific analytic methods applied for its determination. The authors have carried out analyses of sweet wort using the Vren and Wiggall method modified by Lie and Rasch. In the medified method the distorting effects of amino acids are completely eliminated. The modified method is described in detail.(In Czech, English summary only)

Extraction colorimetric method based the biuret reaction can be used to advantage for rough determination of protein content in barley, since its gives reasonably reliable results. Its accuracy depends in the standardization of samples and fineness of grinding. The original method developed by Johnson and Craney has been adapted to meet concrete, specific conditions. Isopropyl alcohol is now used as an extracting agent, basic salt is used instead of cupric carbonate, and vacuum filtration has been replaced with forced pressure membrane filtration. Plants making fodder yeast from synthetic ethanol must use as a rule treated water and it is therefore necessary to prescribe its properties and quality. The authors specify allowable chemical composition, especially the maximum allowable concentration of aluminium and free active chlorine. These two elements are practically always present, since in the conditioning technology chlorine is used as disinfecting agent and aluminium salts for clarification.(In Czech, English summary only)
